“Short-term surrogate biomarkers of chronic lesion expansion.”

To enable predictions of chronic lesion expansion, this study found that chronic brain atrophy (CBA) correlates with future lesion growth. Lesions expand too slowly for practical study in clinical trials, and detecting expansion requires laborious manual analysis. CBA predicted more than 80% of the variance in lesion expansion over the subsequent two years. Mean diffusivity also exhibited some predictive power, while T1 hypointensity was ineffective at predicting lesion expansion. CBA is able to capture damage to axons at the expanding lesion rim, predicting the future extent of the lesion core better than measurements of the existing core.
